While the 1966 24 Hours of Le Mans is forever remembered for Ford's orchestrated 1–2–3 finish, the race featured many other notable contenders at the Circuit de la Sarthe. Among them was Matra, making its debut at the French endurance classic with three examples of its new 620 sports prototype, each powered by a BRM V8 engine.

Only car No.43 retained left-hand drive, and its front right headlight was green to match Henri Pescarolo's helmet. Fresh from his first Formula 3 victories, Pescarolo made his debut at the 1966 24 Hours of Le Mans, replacing Jacky Ickx, who was himself making his first Le Mans appearance in Ford GT40 No.60. He was partnered with Jean-Pierre Jaussaud, who took the start.

An ignition problem forced Jaussaud into the pits after just nine laps. A succession of lengthy pit stops followed, and after completing only 38 laps in eight hours, the team was forced to retire.

The Matra 620s made their final appearance later that year in the 1000km of Paris, where they achieved little of note. Yet, although it was not apparent at the time, Matra had laid the foundations of its Le Mans story. By the end of 1966, all the key figures were in place, and what seemed the end of one chapter was, in reality, the beginning of a remarkable adventure.
